All the ones already mentioned are good (Microsofts being the best), you should also have something like SpywareBlaster to help prevent spyware. Also don't use IE; something like firefox or Opera would be good (I prefer Opera).

Don't even bother with that. Only use it when told so by somebody who knows what they are doing and all other methods have failed. Even if you do have to use Hijackthis, do not remove anything yourself without being told to do so. There are plenty of forums where you can get help with Hijackthis logs and that would be a much better idea.

I remove spyware for the company I work for almost daily through their network, and Spybot Search and Destroy 1.4 has worked better than all the other's I've ever tried, which is a lot.

If the damn employees would just leave the auto-protect features on, we'd be set. But they don't, and we haven't made it password protected yet.

Spyware Doctor detects all the crap my other scanners don't. I recommend that :).

Seconded. Spyware Doctor is very reliable. Having installed it for the first time, it wiped my computer clean of anything useless and the performance of the PC greatly enhanced.

I use a combination of things.

First off switching to Firefox drastically improved how much spyware I get. My scans now rarely turn anything up when I used to get 50+ things each few days. I have protection from SpywareBlaster which helps stop the junk before it even gets in there, and its free. Then I use Ad-aware and Spyware Blaster to finish the job and rid my machine of any that does manager to get in.
